1) The Regulation of Heterologous Subtilin Expression in Bacillus subtilis W168
SUBTILIN: Lantibiotic, Antimicrobial peptide (AMP), Promising alternative for conventional antibotics, Inhibitor of cell wall biosynthesis (G+ bacteria), Natively produced by B. subtilis ATCC6633
OBJECT: This project was developed to express subtilin heterologously by transferring the whole subtilin gene cluster spaBTCSIFEGRK from B. subtilis ATCC6633 into well-established model strain B. subtilis W168 and investigate the underlying regulation of subtilin biosynthesis by the two-component system (TCS) SpaRK and the transcriptional regulators AbrB, Spo0A and SigH (see figure below).
2) Regulatory Network of “Bce-like” Modules in Lactobacillus casei BL23 in Response to Subtilin
OBJECT: This project was developed to explore the interplay of “Bce-like” modules (see figure below) responding to antibiotic subtilin in L. casei BL23, and as well as the regulation of those systems in combination with the dltABCD operon and the mprF gene, both of which are involved in cell wall teichoic acid biosynthesis.
3) Transcriptomic Studies on Cell Envelope Stress Response (CERS) in Bacillus subtilis
OBJECT: This project was developed to comprehensively investigate cell wall stress response in B. subtilis by genome-wide transcriptional profiling using RNA sequencing technique. To reach this aim, we chose 10 different kinds of antibiotics or chemicals that target at important steps of cell wall biosynthesis to treat wild type B. subtilis (see figure below), then followed with RNA sequencing and a series of comparative genomics analyses to get detailed information of the distribution and conservation of cell envelope stress-sensing regulatory systems in B. subtilis.
