Title: Single-cell lineage tracing in development and regeneration
Abstract:
A key goal of biology is to understand the fate choices single cells make during processes such as development or regeneration. Single-cell RNA-sequencing (scRNA- seq) is commonly used to identify cell types in a tissue or organ. However, organizing the resulting taxonomy of cell types into lineage trees to understand the origin of cells remains challenging. Here we present LINNAEUS - a strategy for simultaneous lineage tracing and transcriptome profiling in thousands of single cells to reconstruct lineage trees in zebrafish larvae and the regenerating zebrafish heart. In the latter, we discover transient cell states that shape the regenerative niche. Through a combination of lineage tracing, cell ablation and signaling inhibition, we determine the origin, role and behavior of these transient states in the regenerating heart.
