Description of work
The aim of the subproject A1 is the comprehensive tracking of electron-driven proton translocation in cytochrome c oxidase using time-resolved infrared and resonance Raman spectroscopy. For this purpose, novel techniques are developed and applied, such as the "flow-flash" technique with microfluidic mixer for triggering the reaction by a short laser pulse or electrochemical electron injection into solid-state supported oxidases in combination with surface enhanced infrared and resonance Raman spectroscopy. In this way, the reaction sequence and its speed-determining steps as well as the central amino acids are to be detected.
