Dynamics of hematopoiesis after stem cell transplantation
Our project aims to model the hematopoietic reconstitution dynamics after HSC transplantation by combining high‑resolution barcode tracking with vector‑integration‑site (VIS) analysis. Building on a proven barcode system and corresponding bioinformatic pipelines, our team will generate reference “mini‑bulks,” integrate unique molecular identifiers and compare barcode‑based quantification to state‑of‑the‑art VIS methods. Using a Fanconi‑anemia mouse model, we will test how different conditioning regimens, graft compositions and stress conditions affect the clonal competition between corrected and uncorrected hematopoietic stem cells. These efforts are supported by complementary mathematical modeling of clonal dynamics.
