We compared RTqPCR and RTddPCR for detecting SARS-CoV-2 in wastewater, finding that with proper optimization, the sensitivity and precision gains from using RTddPCR over conventional RTqPCR are marginal. Choosing between the two methods in environmental surveillance depends on available resources and throughput requirements rather than significant performance differences.
