Online monitoring via Flowcytometry for process control of biotechnological synthesis
Recombinant microorganisms play an increasing role for production of biotechnological products. The economic efficiency of this synthesis depends on the maximum production rate depending on the organism and state of physiology. It was shown that in most of the cases the maximum product yield and production rate was not achieved due to a heterogenic population distribution. For rapidly detection of this notwithstanding non optimal behaviour and the adaption of process parameters an online monitoring of the population distribution is necessary. The objective of this project is to achieve basic knowledge about process monitoring and control in biotechnologically relevant processes with recombinant yeasts as an example.
Therefore an online flowcytometry system was used which was developed in cooperation with Cytecs (Görlitz, Germany). Previously the development was funded by the saxon department of environment and agriculture.
The focus is the use of recombinant yeasts with GFP marked protein expression for getting information about the performance of protein synthesis of each individual of the whole population.
