CyTOF®2 (Fluidigm)
The CyTOF2 mass cytometer measures up to 40 parameters on individual cells in suspension that are labeled with stable metal isotope-coupled antibodies. The CyTOF technique is based on inductively coupled plasma (ICP) and time-of-flight (TOF) mass spectrometry to determine properties of cells (cytometry, CyTOF) and was commercialized in 2009.
The advantage over conventional flow cytometry include minimal overlap between metal signals. The relatively low flow rate with approximately 500 cells per second can be overcome by barcoding technique that allows sample pooling and reduction of batch effects.
The high-dimensional mass cytometry data generates FCS files that can be analyzed by FlowJo software, FSC Express or OMIQ. We provide software access and assistance for high-dimensional single cell analysis with FlowSOM, SPADE, CITRUS, and viSNE.
CyTOF XT
4th and newest generation mass cytometer CyTOF XT available at CRTD
We received the new mass cytometer CyTOF XT (Standard BioTools) for automated high-parameter cytometry analysis for basic research and standardized for clinical studies. This unique combination of high-throughput cytometry with time-of-flight mass spectrometry enables single cell proteome profiling of up to 50 markers simultaneously.
